![]() | Staining of Sigma�s Mouse Embryo Multiple Developmental Stages Northern Blot (Product Code BLOT-3) with Blot Stain Blue . Two μg each poly(A)+ mRNA, isolated from Balb\c mouse embryos at 4 different developmental stages (7, 11, 15 and 17 days), were subjected to electrophoresis on a 1.2% Agarose gel in the presence of formaldehyde. After transferred the mRNA to a positively charged nylon membrane and UV crosslinked, the blot was stained with Blot Stain Blue (Product Code B1177) for 2 minutes. The quality of the RNA, e.g. size and amount, as well as the RNA size marker, are well observed by the staining. |

| Reconstitution | 10× Blot Stain Blue should be diluted 1:10 with deionized water prior to use. E.g. Dilute 1 ml of 10× Blot Stain Blue with 9 ml deionized water. |
| Application | Blot Stain Blue is a rapid, reversible, non-interfering staining reagent for detecting RNA immobilized on nylon or nitrocellulose membranes. Assessment of transfer efficiency and visualization of markers is possible without interference with subsequent hybridization procedures. The diluted staining solution is applied to the blot and within only 15 minutes, transferred RNA is stained for easy visualization. Subsequent standard pre-hybridization and hybridization steps reverse the staining and remove the dye from the membrane. Blot Stain Blue is supplied as a 10 × concentrate and should be diluted with water prior to use. |
